Zinc as an inhibitor of NMDA receptor can exhibit antidepressant effect

چکیده
Background : New antidepressant strategies are needed, due to unsatisfactory clinical efficacy and many side effects of commonly used drugs. Recent studies linking the pathophysiology depression with glutamatergic imbalance. There is hyperactivity main excitatory system (glutamatergic) its inhibition (GABAergic). N-methyl D-aspartate (NMDA) receptors as a part synapses potential targets for intervention. Antagonist administration systems, such zinc, can exhibit effects.
Objective To observe effect zinc on NMDA in depressed subjects
Methods In this paper, we provide literature review. The method achieve objective consists using exploration, which was conducted from February June 2022 by searching relevant several databases.
Results Study trials both human animal subjects reveal that associated lower concentration zinc. Comparison between lowest intake highest had significantly incidence developing depression. Dietary deficiency induces along upregulation receptor complexes. Zinc’s might be mediated through action reducing channel-opening frequency.
Conclusions presence may downregulate glutamate response binding receptors. Because numerous about connection depression, it seems have potency develop new antidepressants. Since capability reduce depressive symptoms, expected leading increased medication adherence, costs better outcomes.

Glycine/NMDA Receptor Pathway and Mediating the Rapid-Onset Antidepressant Effect of Alkaloids From Trichilia Monadelpha
Introduction: Major depressive disorder is often associated with suicidal tendencies, and this condition accentuates the need for rapid-acting antidepressants. We previously reported that alkaloids (ALK) from Trichilia monadelpha possess antidepressant action in acute animal models of depression and that this effect is mediated through the monoamine and L-arginine-NO-cGMP pathways. This study i...
متن کاملInteraction of zinc with antidepressants in the forced swimming test in mice.
Recent preclinical data have suggested that glutamate NMDA receptor may be involved in the mechanism of action of antidepressant treatments. Functional antagonists of the NMDA receptor complex exhibit an antidepressant-like effect in animal tests that predict antidepressant activity and in animal models of depression. Zinc, a very potent inhibitor of the NMDA receptor, is active in the forced s...
متن کاملPregnanolone Glutamate, a Novel Use-Dependent NMDA Receptor Inhibitor, Exerts Antidepressant-Like Properties in Animal Models
UNLABELLED A number of studies demonstrated a rapid onset of an antidepressant effect of non-competitive N-methyl-d-aspartic acid receptor (NMDAR) antagonists. Nonetheless, its therapeutic potential is rather limited, due to a high coincidence of negative side-effects. Therefore, the challenge seems to be in the development of NMDAR antagonists displaying antidepressant properties, and at the s...
متن کاملZinc effects on NMDA receptor gating kinetics.
Zinc accumulates in the synaptic vesicles of certain glutamatergic forebrain neurons and modulates neuronal excitability and synaptic plasticity by multiple poorly understood mechanisms. Zinc directly inhibits NMDA-sensitive glutamate-gated channels by two separate mechanisms: high-affinity binding to N-terminal domains of GluN2A subunits reduces channel open probability, and low-affinity volta...
متن کاملEffect of ketamine, an NMDA receptor inhibitor, in acute and chronic orofacial pain.
We examined the analgesic effect of racemic ketamine and its 2 enantiomers in 16 female patients (age: 20-29 years) suffering acute pain after oral surgery and in 7 female patients (age: 42-79 years) suffering chronic neuropathic orofacial pain. All 3 forms of ketamine consistently relieved postoperative pain, (S)-ketamine being 4 times more potent than (R)-ketamine. The analgesic effect was ma...
